Murine Model Crypts

Murine model crypts are the glandular invaginations of mouse intestinal epithelium that house stem and progenitor cells, providing a tractable system for studying tissue renewal and disease. In ex vivo cultures, isolated crypts embed in an extracellular-matrix gel and respond to niche signals such as Wnt, R-spondin, and Noggin, enabling stem cells to generate three-dimensional intestinal organoids with key epithelial cell types. In cancer research, these cultures support analysis of oncogenic mutations, tumor initiation, epithelial regeneration, and responses to candidate therapies under controlled conditions. They also enable comparisons between normal and transformed tissue while complementing whole-animal studies.

In this protocol, we describe how to generate murine primary epithelial colon monolayers directly from intestinal crypts. We provide experimental approaches to generate confluent monolayers on permeable filters, confluent monolayers for scratch wound healing and biochemical studies, and sparse and confluent monolayers for immunofluorescence analysis.
