Organoid Culture Medium

Organoid culture medium is a tailored nutrient solution that supports the growth, organization, and specialized functions of three-dimensional cell models in vitro. It combines basal nutrients with growth factors, signaling molecules, and other supplements that recreate key aspects of a tissue-specific cellular niche, guiding cell survival, proliferation, differentiation, and polarity within an extracellular matrix scaffold. In cancer research, optimized media help establish and maintain patient-derived tumor organoids from biopsies or surgical samples. These models can preserve important features of the original tumor and support studies of cancer biology, treatment response, resistance, and individualized drug selection.

Using a laminin-rich extracellular matrix as a scaffold and culture media for nutrients, cells are facilitated to multiply and self-organize within the organoid. A simple method to establish primary murine colon tumor organoid is described. This method utilizes the feature that colon tumor cells survive and grow into organoids in media containing limited growth factors, whereas normal colon epithelial do not.
