Patient-derived Culture

Patient-derived culture is an ex vivo model in which cells or tissue collected from an individual are maintained under controlled laboratory conditions, enabling cancer research that reflects patient-specific biology. Tumor material is processed, placed in a suitable culture environment, and supported with nutrients and growth signals so viable cells can proliferate while retaining some features of the original sample. These cultures allow researchers to examine tumor growth, cellular behavior, therapeutic sensitivity, and resistance mechanisms. They support disease modeling, drug screening, biomarker evaluation, and studies of treatment response, helping connect experimental findings with personalized cancer care.

This article describes the preparation of freshly obtained melanoma tissue into primary cell cultures, and how to remove contaminations of erythrocytes and fibroblasts from the tumor cells. Finally, we describe how CD133+ putative melanoma stem cells are sorted from the CD133- bulk using Magnetic Activated Cell Sorting (MACS).

The following protocol focuses on the establishment of a primary culture of patient-derived soft tissue sarcoma (STS). This model could help us to better understand the molecular background and pharmacological profile of these rare malignancies and could represent a starting point for further research aimed at improving STS management.
