Cancer Cell Dormancy

Cancer cell dormancy is a reversible state in which viable tumor cells pause proliferation, allowing them to persist undetected after treatment or before forming a detectable metastasis. Dormant cells commonly exhibit cell-cycle arrest and altered metabolism, while signals from the tissue microenvironment, immune system, and surrounding extracellular matrix can maintain or release dormancy; changes in these conditions may reactivate growth. In cancer research, studying dormancy helps explain late recurrence and metastatic relapse, identify vulnerabilities that differ from those of rapidly dividing tumors, and guide strategies for detecting, eliminating, or permanently restraining residual disease.

An In Vitro Dormancy Model of Estrogen-sensitive Breast Cancer in the Bone Marrow: A Tool for Molecular Mechanism Studies and Hypothesis Generation

We developed an in vitro model of dormancy in the bone marrow for estrogen-sensitive breast cancer cells. The goal of this protocol is to demonstrate use of the model for the study of the molecular and cellular biology of dormancy and for generation of hypotheses for subsequent testing in vivo.

A modified 3-D in vitro system is presented in which growth characteristics of several tumor cell lines in reconstituted basement membrane correlate with the dormant or proliferative behavior of the tumor cells at a metastatic secondary site in vivo.
