Dialysate

Dialysate is a carefully formulated fluid used in dialysis to remove metabolic waste and correct electrolyte and acid-base imbalances when the kidneys cannot maintain internal chemical balance. During hemodialysis, it flows on one side of a semipermeable membrane while blood flows on the other, allowing urea, excess electrolytes, and other small solutes to move by diffusion; pressure-driven ultrafiltration removes excess water. In peritoneal dialysis, the solution enters the abdominal cavity and uses the peritoneal membrane for exchange, with glucose or another osmotic agent drawing fluid from the blood. Dialysate composition and flow strongly influence treatment safety, efficiency, and patient outcomes.

Dialysis: Diffusion Based Separation

2023

Dialysis is a common technique used in biochemistry for separating molecules based on diffusion. In this procedure, a semipermeable membrane allows the movement of certain molecules based on size. This method can be applied to the removal of buffer, known as desalting, or exchanging buffer molecules or ions from a protein solution. This video covers the principles of dialysis along with a general procedure. Several applications of dialysis are reviewed, including the removal of gradient...

Microdialysis of Ethanol During Operant Ethanol Self-administration and Ethanol Determination by Gas Chromatography

2012

A method to determine the time course of ethanol concentration in the brains of rats during operant ethanol self-administration is described. Gas chromatography with flame ionization detection is used to quantify ethanol in the dialysate samples, because it has the sensitivity required for the small volumes that are generated.

Source: Soukupová, M., et al. Microdialysis of Excitatory Amino Acids During EEG Recordings in Freely Moving Rats. J. Vis. Exp. (2018)This video demonstrates the assessment of extracellular excitatory neurotransmitter levels in rats with chronic epilepsy induction. After implanting a microdialysis probe into the brain and confirming the absence of seizures using electroencephalography (EEG), extracellular fluid is collected to evaluate elevated neurotransmitter levels in epileptic rats. A...

Protein transduction enables the direct delivery of biologically active proteins into cells. In contrast to conventional methods such as DNA transfection or viral transduction this non-invasive paradigm allows highly efficient cellular manipulation in a titratable manner circumventing cellular toxicity and the risk of oncogenic transformation by permanent genetic modification.
