IgE-mediated responses are immune reactions in which immunoglobulin E (IgE) antibodies drive inflammation, most commonly in allergic disease, making them important in clinical diagnosis and care. After sensitization, an allergen cross-links IgE bound to high-affinity FcεRI receptors on mast cells and basophils, triggering degranulation and release of histamine, leukotrienes, and other mediators. These substances can produce rapid symptoms such as itching, wheezing, urticaria, or anaphylaxis, while later inflammatory signaling may prolong tissue effects. Understanding this mechanism supports allergy testing, risk assessment, and treatments that reduce mediator release or interrupt IgE-dependent activation.

Basophil activation test is a potent tool for the detection of IgE-dependent allergies in vitro. Here, an optimized protocol for basophil activation test is used to investigate drug hypersensitivity. A method for the efficient production of covalent drug-protein conjugates and their physicochemical characterization is described.

In vitro analysis of class switch recombination in mice is challenging due to cytophilic IgE molecules bound to Fc receptors on the surface of B cells. We describe a method for IgE detection using trypsin-mediated cleavage of surface-bound IgE prior to fixation and permeabilization for cytoplasmic fluorescence staining.

In this video, we describe an in vitro method to evaluate the allergic potential of an antigen to induce basophil degranulation. β-hexosaminidase is used as an index of chemical mediator release, and the enzyme activity of β-hexosaminidase is measured using a fluorogenic substrate.

The current study describes the development and applications of a genetically engineered assay system based on the transfection of rat basophilic leukemia cells with the equine FcεRIα gene. Transfected cells express a functional receptor where the release of mediators of the allergic response can be activated by IgE and antigen.
