Human Amniotic Epithelial Cells

Human amniotic epithelial cells are specialized cells lining the inner surface of the amniotic membrane, where they help maintain the fetal environment during development. Their biological activity reflects epithelial barrier functions and the release of signaling molecules, while their capacity for plasticity allows them to respond to developmental cues and adopt characteristics of other cell types under defined conditions. In developmental biology, these cells provide a model for studying amnion formation, cell differentiation, and interactions between fetal tissues and their surrounding membranes. Their accessibility and low immunogenicity also support research into tissue repair, cell-based therapies, and regenerative medicine.

Here we describe a detailed method for growing primary human bronchial epithelial cells from explants of human bronchial airway tissue including differentiated growth on an air-liquid interface. The fallopian tube (FT) is emerging as an alternative site of origin for serous ovarian carcinoma (SOC). This protocol describes a novel method for the isolation and ex vivo culture of fallopian tube epithelial cells. This system recapitulates the in vivo epithelium and allows the study of SOC pathogenesis.
