Progenitor Subset Isolation

Progenitor subset isolation is the process of separating developmentally relevant progenitor cells from a heterogeneous population so their properties and lineage potential can be studied. Researchers typically identify distinguishing cell-surface markers, label the cells with specific antibodies, and enrich the target population using methods such as flow cytometric sorting or magnetic-activated cell separation. Isolated subsets can then be evaluated for self-renewal, differentiation, gene expression, and responses to developmental signals. In developmental biology, this approach helps clarify how progenitors generate specialized cell types, supports studies of tissue formation and regeneration, and improves the reproducibility of organoid, disease-modeling, and cell-based research.

Isolation and Enrichment of Liver Progenitor Subsets Identified by a Novel Surface Marker Combination

Liver injuries are accompanied by progenitor cell expansion that represents a heterogeneous cell population. Novel classification of this cellular compartment allows for the distinguishing of multiple subsets. The method described here illustrates the flow cytometry analysis and high purity isolation of various subsets that can be used for further assays.

Isolation of Precursor B-cell Subsets from Umbilical Cord Blood

Here we describe a protocol for isolating subsets of precursor B-cells from umbilical cord blood. A sufficient quantity and quality of nucleic acids may be extracted from the cells and used in subsequent assays utilizing DNA or RNA.

Distinct dendritic cell subsets exist as rare populations in lymphoid organs, and therefore are challenging to isolate in sufficient numbers and purity for immunological experiments. Here we describe a high efficiency, high yield method for isolation of all of the currently known major subsets of mouse splenic dendritic cells.

Naïve CD4+ T cells polarize to various subsets depending on the environment at the time of activation. The differentiation of naïve CD4+ T cells to various effector subsets can be achieved in vitro through the addition of T cell receptor stimuli and specific cytokine signals.

A method to analyse the distribution of bone marrow hematopoietic progenitors in flow cytometry as well as to efficiently isolate highly purified hematopoietic stem cells (HSCs) is described. The isolation procedure is essentially based on magnetic enrichment of c-Kit+ cells and cell sorting to purify HSCs for cellular and molecular studies.
