X-ray Micro-computed Tomography

X-ray micro-computed tomography (micro-CT) is a nondestructive imaging technique that produces three-dimensional maps of a specimen’s internal structure at micrometer-scale resolution. During scanning, an object rotates through an X-ray beam while a detector records projections; computational reconstruction then converts differences in X-ray attenuation into a volumetric image composed of voxels. In engineering, micro-CT reveals pores, cracks, inclusions, fibers, and other features without cutting or otherwise damaging components. It supports material characterization, additive-manufacturing quality control, failure analysis, dimensional inspection, and validation of computational models, helping researchers link internal architecture to mechanical performance and manufacturing processes.

Here we show how to process tree cores with an X-ray computed tomography toolchain. Except for chemical extraction for some purposes, no further physical lab treatment is needed. The toolchain can be used for biomass estimations, for obtaining MXD/tree-ring width data as well as for obtaining quantitative wood anatomy data.

Source: Kim, D., et. al. Combined Near-infrared Fluorescent Imaging and Micro-computed Tomography for Directly Visualizing Cerebral Thromboemboli. J. Vis. Exp. (2016)This video demonstrates visualization of cerebral thrombus in mice using micro-computed tomography (micro-CT) imaging. A cerebral thrombus mouse model is injected with fibrin-targeted gold nanoparticles acting as a contrasting agent. The mouse is placed on the bed of a micro-CT machine, and images are acquired from multiple angles.

Micro X-ray computed tomography is effective in obtaining three-dimensional information from undamaged human specimens but has limited success in observing soft tissues. The use of phosphotungstic acid contrast agent can resolve this issue. We implemented this contrast agent to examine human delicate fibromuscular tissues (the orbicularis retaining ligament).
