Hot Water Extraction

Hot water extraction is a sample-preparation and separation technique that uses heated water to transfer soluble compounds from a solid or complex matrix into an aqueous extract. By raising temperature, water can improve compound solubility and molecular diffusion, while contact time and mixing influence how efficiently target substances enter the liquid phase. In environmental research, the method can support analysis of soils, sediments, and other materials for water-soluble chemicals, helping researchers characterize contamination and evaluate treatment or recovery processes. Its use of water rather than organic solvents can also reduce hazardous solvent waste, although extraction selectivity depends on the sample and target compounds.

Employing Pressurized Hot Water Extraction (PHWE) to Explore Natural Products Chemistry in the Undergraduate Laboratory

Here, we employ a pressurized hot water extraction (PHWE) method, which utilizes an unmodified household espresso machine to introduce undergraduates to natural products chemistry in the laboratory. Two experiments are presented: PHWE of eugenol and acetyleugenol from cloves and PHWE of seselin and (+)-epoxysuberosin from the Australian plant Correa reflexa.

Purification and Visualization of Lipopolysaccharide from Gram-negative Bacteria by Hot Aqueous-phenol Extraction

We describe a modified hot aqueous-phenol extraction method for purifying lipopolysaccharide (LPS) from Gram-negative bacteria. Once extracted, the LPS can be subsequently analyzed by SDS-PAGE and visualized by direct staining or Western immunoblot.

Determining Viral Disinfection Efficacy of Hot Water Laundering

In response to the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) pandemic, a laboratory protocol was developed to test the viral disinfection efficacy of hot water laundering of cloth face coverings, cotton scrubs, and denim pants. The Phi6 virus (bacteriophage) was used as the organism to test disinfection efficacy.

This protocol presents a fully automated workflow for the extraction of geosmin and 2-methylisoborneol from water and lipid-rich fish tissues. The method allows for the early detection of these molecules before they reach odor thresholds. Representative data from an aquaculture setting are provided.
