Hplc-icp-ms Analysis

HPLC-ICP-MS analysis is a hyphenated analytical technique that separates chemical species by high-performance liquid chromatography and then measures their elemental composition with inductively coupled plasma mass spectrometry. In operation, HPLC resolves compounds according to interactions with the stationary and mobile phases, while the eluent is nebulized into an argon plasma, where analytes are atomized and ionized before ions are sorted by mass-to-charge ratio. This combination provides sensitive, element-specific detection and distinguishes different forms of the same element. In environmental research, it supports trace-metal speciation in water, soil, sediment, and biological samples, helping assess contaminant transport, exposure, toxicity, and treatment outcomes.

Here, we describe in great detail an established and robust protocol for the extraction of glucosinolates from ground plant materials. After an on-column sulfatase treatment of the extracts, the desulfoglucosinolates are eluted and analyzed by high-pressure liquid chromatography.

This study developed a high-performance liquid chromatography coupled with triple quadrupole mass spectrometry (HPLC-QQQ-MS) method to identify and quantify tropane alkaloids (hyoscyamine, anisodamine, scopolamine) in Anisodus tanguticus. The method proved rapid, accurate, and reliable for high-throughput analysis of medicinal plants.
