Serial Block Face Imaging

Serial block face imaging is a three-dimensional microscopy technique that reveals the internal architecture of a specimen by repeatedly imaging its surface as successive layers are removed. In a scanning electron microscope, an ultramicrotome or focused cutting system removes a thin section from the exposed block face, and automated image acquisition records each newly revealed surface for computational alignment and reconstruction. Applied to environmental research, the method can characterize complex microstructures in soil, sediments, biofilms, and other heterogeneous materials at high spatial resolution. These three-dimensional datasets help link physical organization with transport pathways, microbial habitats, and material transformations that are difficult to resolve in conventional two-dimensional images.
