Live Dead Discrimination

Live-dead discrimination is the classification of cells or microorganisms as viable or nonviable, a distinction that helps researchers assess biological activity and environmental health. In common fluorescence-based assays, membrane-permeable dyes label cells with intact membranes, while membrane-impermeable dyes enter cells with damaged membranes and produce a contrasting signal; some methods also detect metabolic activity. Environmental scientists use these approaches to evaluate microbial communities in water, soil, sediments, and biofilms, including responses to pollutants or treatment processes. The resulting viability profiles complement cell counts by indicating whether detected organisms are likely active, injured, or no longer viable.

This manuscript describes a protocol to examine the olfactory system of rodents. The olfactory habituation/dishabituation test will allow investigators to determine whether a mouse habituates to a repeatedly presented odor and whether the mouse demonstrates dishabituation when presented a novel odor.

Here, we present a protocol to quantify post-stress survival in yeast samples. The assay employs two fluorescent dyes, SYTO 9 and propidium iodide (PI), to quantify plasma membrane integrity. It uses flow cytometry to provide quantitative and reproducible estimates of the live, dead, and damaged cell fractions for post-oxidative-stress samples.
