Adapter Ligation Inhibition

Adapter ligation inhibition is a molecular biology strategy that prevents sequencing adapters from joining DNA or RNA fragments at undesired sites during genetic library preparation. It works by making nucleic acid ends nonreactive, for example through removal or blocking of 5′ phosphate groups, end modification, or complementary blocking oligonucleotides that limit access to ligation substrates. By suppressing adapter self-ligation and nonspecific joining, this approach reduces adapter dimers and unwanted library products, increasing the proportion of sequenceable molecules. In genetics, controlled ligation inhibition supports more efficient next-generation sequencing workflows, improves library complexity, and can enhance the accuracy of downstream variant detection and genomic analysis.

DNA Ligation Reactions

In molecular biology, ligation refers to the joining of two DNA fragments through the formation of a phosphodiester bond. An enzyme known as a ligase catalyzes the ligation reaction. In the cell, ligases repair single and double strand breaks that occur during DNA replication. In the laboratory, DNA ligase is used during molecular cloning to join DNA fragments of inserts with vectors – carrier DNA molecules that will replicate target fragments in host organisms. The surgical procedure used to induce experimental myocardial infarction in mice begins with left thoracotomy between the third and the fourth ribs in order to visualize the anterior surface of the heart and left lung. The left coronary artery is ligated, the chest is closed and the mouse is allowed to recover spontaneously.

Here we show how to use Proximity Ligation Assay (PLA), with a combination of antibodies to visualize Bone Morphogenetic Protein (BMP) signaling in fixed cells. This technique allowed us to follow the nuclear accumulation of endogenous BMP activated effector-complexes and quantify their levels over time under BMP4 stimulation.
