Carnoy's Fixative

Carnoy’s fixative is a rapid, alcohol-based preservative used to stabilize cells and tissues for microscopic and cytogenetic analysis. Typically composed of methanol and glacial acetic acid, it penetrates specimens quickly, precipitates proteins, removes lipids, and helps preserve the size, shape, and optical contrast of chromosomes. In genetics, researchers use Carnoy’s fixative during chromosome preparation, including cell harvesting and slide-making for karyotyping, mitotic chromosome spreads, and analysis of chromosomal number or structure. Its rapid action and compatibility with staining methods make it valuable for examining dividing cells, although fixation conditions must be controlled to minimize chromosome distortion and loss of cellular detail.
