Cholinergic Anti-inflammatory Pathway

The cholinergic anti-inflammatory pathway is a neuroimmune communication system that limits excessive inflammation through cholinergic signaling. In this pathway, vagus nerve activity promotes the release of acetylcholine, which binds nicotinic acetylcholine receptors, particularly α7 nicotinic receptors, on macrophages and other immune cells to reduce the production of proinflammatory cytokines such as tumor necrosis factor. This mechanism helps regulate host responses during infection, tissue injury, and systemic inflammation while preserving essential immune functions. Understanding the pathway supports research into neuroimmune interactions and potential treatments for inflammatory and infectious diseases, including pharmacological and bioelectronic approaches that modulate vagal signaling.
