Dual Promoter-reporter System

A dual promoter-reporter system is a molecular biology method that measures the activity of two promoters in the same experimental setting, helping researchers compare gene regulation under identical conditions. In this approach, each promoter controls a different reporter gene, and the resulting signals are quantified to assess promoter-specific transcription, regulatory responses, or normalization controls. In immunology and infection research, the system can reveal how host or pathogen factors influence immune-related promoters, inflammatory signaling, and virulence-associated gene expression. By distinguishing parallel regulatory outputs, it supports mechanistic studies of infection, screening of regulatory factors, and evaluation of therapeutic strategies.

Promoter expression analyses are crucial to improving the understanding of gene regulation and the spatiotemporal expression of target genes. Herein we present a protocol to identify, isolate, and clone a plant promoter. Further, we describe the characterization of the nodule-specific promoter in the common bean hairy roots.
