Epitope Conservation Analysis

Epitope conservation analysis is a bioinformatic method that identifies immune-recognized regions of a pathogen that remain similar across strains, variants, or related species. It compares protein or genome sequences, maps predicted or experimentally defined B-cell and T-cell epitopes, and measures sequence conservation to distinguish stable targets from highly variable regions. In immunology and infection research, this analysis helps prioritize epitopes for broadly protective vaccine design, evaluate whether diagnostic targets remain reliable as pathogens evolve, and interpret how mutations may alter immune recognition. By linking sequence variation with antigenic stability, it supports surveillance and the development of strategies that address pathogen diversity.

Conservation of Energy Approach to System Analysis

2023

Source: Ricardo Mejia-Alvarez and Hussam Hikmat Jabbar, Department of Mechanical Engineering, Michigan State University, East Lansing, MI The purpose of this experiment is to demonstrate the application of the energy conservation equation to determine the performance of a flow system. To this end, the energy equation for steady, incompressible flow is applied to a short pipe with a gate valve. The gate valve is then gradually closed and its influence on flow conditions is characterized. In...

2014

Biochemical assays with recombinant human MHC II molecules can provide rapid, quantitative insights into immunogenic epitope identification, deletion, or design. Here, a peptide-MHC II binding assay scaled to 384-well plates is described. This cost effective format should prove useful in the fields of protein deimmunization and vaccine design and development.

This protocol describes the use of peptide:MHC tetramers and magnetic microbeads to isolate low frequency populations of epitope-specific T cells and analyze them by flow cytometry. This method enables the direct study of endogenous T cell populations of interest from in vivo experimental systems.

Conservation of Momentum

Source: Nicholas Timmons, Asantha Cooray, PhD, Department of Physics & Astronomy, School of Physical Sciences, University of California, Irvine, CA The goal of this experiment is to test the concept of the conservation of momentum. By setting up a surface with very little friction, collisions between moving objects can be studied, including their initial and final momenta. The conservation of momentum is one of the most important laws in physics. When something is conserved in physics, the...

Conserved Binding Sites

2020

Many proteins’ biological role depends on their interactions with their ligands, small molecules that bind to specific locations on the protein known as ligand-binding sites. Ligand-binding sites are often conserved among homologous proteins as these sites are critical for protein function. Binding sites are often located in large pockets, and if their location on a protein’s surface is unknown, it can be predicted using various approaches. The energetic method computationally analyses the...
