Hemagglutinin Detection

Hemagglutinin detection is the identification or measurement of hemagglutinin, a surface glycoprotein that enables many viruses, including influenza viruses, to attach to host cells and initiate infection. In a hemagglutination assay, hemagglutinin binds sialic acid-containing receptors on red blood cells, producing a visible lattice rather than a compact cell pellet; related inhibition and immunoassays detect blocking antibodies or the protein itself. These methods support viral identification, antigen quantification, vaccine evaluation, and surveillance of antigenic change. In immunology and infection research, hemagglutinin detection links molecular recognition with infectivity and helps assess immune protection and emerging viral variants.

Here we describe a way to express correctly folded and functional influenza virus surface antigens derived from the novel Chinese H7N9 virus in insect cells. The technique can be adapted to express ectodomains of any viral or cellular surface proteins.

We describe a method to measure the activation of Fc-mediated effector functions by antibodies that target the influenza virus hemagglutinin. This assay can also be adapted to assess the ability of monoclonal antibodies or polyclonal sera targeting other viral surface glycoproteins to induce Fc-mediated immunity.

A simple slot blot method was developed for the quantification of influenza viral hemagglutinin and neuraminidase using universal antibodies targeting their most conserved sequences identified through bioinformatics analyses. This innovative approach may provide a useful alternative to quantitative determination of all viral hemagglutinin and neuraminidase.
