Intestinal Manipulation

Intestinal manipulation is the deliberate alteration of the gut’s physical, microbial, chemical, or immune environment to investigate how the intestine functions and responds to disease. In immunology and infection research, interventions such as changing microbial communities, epithelial barrier conditions, or immune signaling can modify host–microbe interactions through effects on pattern-recognition pathways, cytokine production, and intestinal permeability. These approaches help researchers distinguish how commensal organisms, pathogens, and host tissues contribute to inflammation, colonization, and systemic immune responses. Intestinal manipulation supports studies of infection mechanisms, microbiome–immune crosstalk, inflammatory disease, and the development of targeted therapies.

Postoperative ileus (POI) is a complication of abdominal surgery leading to increased morbidity and a prolonged hospital stay. Because prophylactic or therapeutic strategies are lacking intensified research is necessary. Therefore we established a standardized and feasible mouse model to investigate the pathophysiology of POI and to study potential therapeutic options.

Generation and Manipulation of Rat Intestinal Organoids

Here, we present a protocol to generate rat intestinal organoids and use them in several downstream applications. Rats are often a preferred preclinical model, and the robust intestinal organoid system fills the need for an in vitro system to accompany in vivo studies.

In this video, we demonstrate a technique to isolate whole crypts from a mouse’s small intestine. The isolated whole crypts can be used to propagate long-lived and self-renewing 3D organoids in vitro.
