Macrophage Death Detection

Macrophage death detection is the assessment of whether macrophages remain viable or undergo apoptosis, necrosis, or other forms of regulated cell death, an important readout in immunology and infection research. It works by combining changes in cell morphology and membrane integrity with biochemical or fluorescent markers, such as loss of metabolic activity, membrane permeability, nuclear fragmentation, or activation of death-associated pathways, to distinguish dying cells from healthy ones. These measurements help determine how pathogens, toxins, inflammatory signals, or experimental treatments affect macrophage survival and function, clarifying host defense, tissue damage, and potential therapeutic responses.

We describe the detection of NLRP3 inflammasome activation on cellular basis using fluorescence microscopy and staining for active caspase-1 and the adaptor, ASC. A lactate dehydrogenase release assay is presented to detect pyroptotic lysis on a population basis. These techniques can be adapted to study many aspects of inflammasome biology.
