Opsonized Biofilm Assay

An opsonized biofilm assay is an experimental method for assessing how immune factors alter interactions between phagocytic cells and microorganisms embedded in a biofilm. During the assay, biofilms are exposed to opsonins, such as antibodies or complement components, which bind microbial surfaces and enhance recognition and uptake by immune cells; researchers then quantify measures such as cell association, engulfment, or microbial survival. This approach helps characterize host-pathogen interactions, evaluate immune clearance of surface-associated infections, and compare how biofilm formation or treatment affects susceptibility to immune attack.

This video demonstrates an opsono-adherence assay for examining the interaction between opsonized bacteria and immune cells. Fluorescently labeled bacteria, treated with test serum containing anti-capsule antibodies, are incubated with macrophages. After staining the macrophages with fluorescence, the attached bacteria on the macrophage surface are observed under a microscope for visualization.
