Mesenteric Lymph Duct Cannulation

Mesenteric lymph duct cannulation is a surgical technique for collecting lymph that drains the intestine, providing direct access to immune cells, antigens, lipids, and microbial products transported from the gut. A catheter is placed in a mesenteric lymphatic vessel, allowing lymph to be collected over time and analyzed for cellular, molecular, and soluble components. In immunology and infection research, this method helps characterize intestinal immune trafficking, antigen transport, inflammatory responses, and host interactions with enteric pathogens. By linking gut events to lymphatic dissemination, it supports studies of mucosal immunity, vaccine responses, and disease mechanisms that may be missed through blood sampling alone.

Here we describe a technique to cannulate the mesenteric lymph duct in rats which enables quantification of lipid and drug transport via the lymphatic system following intestinal delivery. The technique can be adapted to assess mesenteric lymph concentrations and/or transport of fluid, immune cells, peptides, proteins and lipophilic molecules.

Cannulation of the Mouse Submandibular Salivary Gland via the Wharton's Duct

A protocol for the cannulation of the mouse submandibular salivary gland via the Wharton's duct is described. For this experiment, the trypan blue solution is used as a dyer to demonstrate how this technique effectively delivers infusions into the targeted gland, and to suggest the reliability of this new approach as a potential clinical drug/cell therapy for the regeneration of salivary glands.

Here, we describe a minimally invasive laparoscopic technique for serial sampling of liver and mesenteric lymph nodes (MLN) in macaques that allows for increased sampling frequency, and reduces the potential for surgical complications when compared to performing a laparotomy.

This manuscript provides a detailed protocol to assess in isolated segments of small mesenteric arteries from rats, vasoactivity and the effect of intraluminal pressure on vascular diameter.
