Nitroblue Tetrazolium

Nitroblue tetrazolium (NBT) is a redox-sensitive dye used to detect cellular reducing activity, especially the oxidative burst of phagocytic immune cells during antimicrobial defense. When neutrophils or other phagocytes generate superoxide through NADPH oxidase, NBT is reduced to an insoluble blue formazan product, allowing respiratory-burst activity to be assessed microscopically or spectrophotometrically. The NBT reduction test has supported the evaluation of phagocyte function and the investigation of defects such as chronic granulomatous disease, in which impaired reactive oxygen species production compromises microbial killing. NBT-based assays remain valuable in immunology and infection research for linking cellular mechanisms to host susceptibility.

A 96-well microtiter plate-based protocol using a 2,3-bis(2-methoxy-4-nitro-5-sulfophenyl)-5-carboxanilide-2H-tetrazolium (XTT) reduction assay is described herein, to study antibodies' effects on biofilms formed by C. tropicalis. This in vitro protocol can be used to check the effect of potential new antifungal compounds on the metabolic activity of Candida species cells in biofilms.
