Gamma-delta T-cell-deficient Mice

Gamma-delta T-cell-deficient mice are experimental animals genetically altered to lack γδ T lymphocytes, allowing researchers to examine their contribution to immune defense and disease. In common models, disruption of the Tcrd gene blocks formation of the T-cell receptor required for γδ T-cell development, while leaving αβ T-cell compartments largely intact. These mice help distinguish γδ T-cell functions in epithelial surveillance, cytokine production, inflammation, and responses to bacterial, viral, or parasitic infection. Comparing deficient and control animals can reveal whether γδ T cells protect against pathogens, worsen immunopathology, or shape adaptive immunity, informing infection biology and immunotherapeutic research.

We describe here an isolation method to obtain human endocervical intraepithelial lymphocytes for the analysis of intraepithelial gamma delta T cells. This protocol can be extended for the purification of endocervical gamma delta T cells by magnetic beads or by cell sorting.

Expansion and Enrichment of Gamma-Delta (γδ) T Cells from Apheresed Human Product

Presented is a protocol for the expansion of Gamma-Delta (γδ) T cell drug product. Lymphocytes are isolated by elutriation and γδ-enriched with zoledronic acid and interleukin-2. Alpha-beta T cells are depleted using a clinical-grade magnetic separation device. The γδ cells are co-cultured with K562-derived, artificial antigen-presenting cells and expanded.

A method is described to propagate human retinoblastoma tumors in mice. Tumor cells are directly injected into the eyes of immune deficient mice. Secondary tumors have been successfully established using both cells directly harvested from human tumors and cultured tumorspheres.
