Organelle Remodeling

Organelle remodeling is the regulated alteration of organelle structure, distribution, composition, or activity in response to cellular signals, enabling cells to adapt to changing conditions. In immunology and infection, signaling pathways coordinate processes such as mitochondrial fission and fusion, lysosomal trafficking, membrane contact formation, and cytoskeletal transport to reshape intracellular compartments. These changes can regulate energy production, inflammatory signaling, antigen processing, and the delivery of antimicrobial components. Pathogens may also manipulate organelle dynamics to create replication-supportive niches or evade host defenses, making organelle remodeling an important focus for understanding infection mechanisms and identifying therapeutic targets.

Herein we describe two approaches to characterize cell polarization events in B lymphocytes during the formation of an IS. The first, involves quantification of organelle recruitment and cytoskeleton rearrangements at the synaptic membrane. The second is a biochemical approach, to characterize changes in composition of the centrosome, which undergoes polarization to the immune synapse.
