Peyer's Patches

Peyer’s patches are organized lymphoid tissues in the small intestine that monitor intestinal contents and coordinate mucosal immune defense. Specialized M cells transport luminal antigens across the epithelium, allowing dendritic cells and other antigen-presenting cells to activate T and B lymphocytes within lymphoid follicles; B cells can then support production of IgA that protects the intestinal surface. In immunology and infection research, Peyer’s patches provide a key site for studying pathogen surveillance, oral antigen responses, immune tolerance, and interactions between the gut microbiota and host immunity. Their function helps explain how the intestine responds to harmful microbes while limiting unnecessary inflammation against food and resident microorganisms.

Here, we describe a precise method for collecting thoracic duct lymphocytes and observing the migration of gut-tropic lymphocytes in rat Peyer’s patches for 3 hours using time-lapse photography. This technique can clarify how the dynamics of lymphocytes are affected under inflammatory conditions.
