Phagocytosis Colony Counting

Phagocytosis colony counting is a microbiological assay that measures the number of viable bacteria engulfed and retained by phagocytic immune cells, providing a quantitative view of host-pathogen interactions. In a typical workflow, immune cells are incubated with bacteria, extracellular organisms are removed or inactivated, and the cells are lysed to release internalized bacteria for serial dilution and colony-forming unit (CFU) enumeration. The resulting counts indicate phagocytic uptake and intracellular bacterial survival under defined experimental conditions. In immunology and infection research, this method helps compare immune-cell function, bacterial persistence, and the effects of antimicrobial treatments or genetic changes on infection outcomes.

We describe methods for live-cell video microscopy of Candida albicans phagocytosis by macrophages. These methods enable stage-specific analysis of macrophage migration, recognition, engulfment and phagosome maturation and reveal novel aspects of phagocytosis.
