Semen Instillation

Semen instillation is a research procedure that introduces semen or seminal fluid into the reproductive tract to examine its effects on mucosal tissues, immune responses, or infection transmission. The method exposes epithelial and immune cells to sperm, seminal plasma proteins, cytokines, and, when experimentally included, infectious agents, allowing investigators to assess local inflammation, immune modulation, and pathogen movement across the mucosal barrier. In immunology and infection research, semen instillation helps model sexual exposure, investigate how seminal factors influence susceptibility or resistance to infection, and evaluate host responses relevant to sexually transmitted pathogens. Controlled administration also supports reproducible studies of reproductive-tract biology and mucosal defense.

Intratracheal instillations deliver solutes directly into the lungs. This procedure targets the delivery of the instillate into the distal regions of the lung, and is therefore often incorporated in studies aimed at studying alveoli. We provide a detailed survival protocol for performing intratracheal instillations in mice.

A Simple Microaspiration Technique for Isolating Somatic Cells from Cryopreserved Equine Semen as Nuclear Donors for Cloning

2025

This protocol presents a simple microaspiration technique to isolate viable somatic cells (SCs) from cryopreserved equine semen. Unlike traditional approaches, this technique avoids long in vitro cultures and overcomes sperm contamination, enabling immediate use in somatic cell cloning.

Intrabronchial Instillation of Streptococcus pneumoniae in Immunocompetent Rodent Models
