Syngeneic Mouse Model

A syngeneic mouse model is an experimental system in which cells or tissues are transferred between genetically identical or closely matched animals, allowing researchers to study biology in an immunocompetent host. Because donor and recipient share the same genetic background, the graft avoids major histocompatibility barriers while preserving immune recognition of foreign, diseased, or altered cells. In immunology and infection research, these models help clarify host–pathogen interactions, inflammatory responses, immune regulation, and treatment effects under physiologically relevant conditions. They are especially valuable for testing vaccines, immunotherapies, and infection-control strategies while accounting for interactions between pathogens, tissues, and an intact immune system.

Syngeneic mouse orthotopic allografts of pancreatic ductal adenocarcinoma (PDAC) recapitulate the biology, phenotypes, and therapeutic responses of disease subtypes. Owing to their fast, reproducible tumor progression, they are widely used in preclinical studies. Here, we show common practices to generate these models, injecting syngeneic murine PDAC cultures into the pancreas.

Implementation of an orthotopic model of renal cell carcinoma in immunocompetent mice affords the investigator a clinically-relevant system defined by the presence of a primary renal tumor and lung metastases in the same animal. This system can be used to preclinically test a variety of treatments in vivo.
