Target Cell Labeling

Target cell labeling is the process of marking cells that are recognized or attacked by immune effectors, enabling their identification, tracking, or measurement during immune responses. Labels may include fluorescent dyes, antibody-linked probes, or radioactive tracers that associate with target-cell membranes or intracellular components, allowing researchers to distinguish target cells from lymphocytes and detect changes such as death, uptake, or loss of membrane integrity. In immunology and infection research, labeled target cells support cytotoxicity assays, flow cytometry, and microscopy to quantify natural killer cell or cytotoxic T-cell activity, evaluate pathogen-induced damage, and compare immune responses across experimental conditions.

Targeted cell delivery is useful in a variety of biomedical applications. The goal of this protocol is to use superparamagnetic iron oxide nanoparticles (SPION) to label cells and thereby enable magnetic cell targeting approaches for a high degree of control over cell delivery and localization.

In this video, we demonstrate the procedure to determine the helicase activity of a target protein to unwind the biotin-labeled dsRNA substrate. The activity of the enzyme was identified by analyzing the electrophoretic mobility shift, followed by a chemiluminescence assay using chemiluminescent enzyme-conjugated streptavidin.
