Acetaminophen Hepatitis Model

An acetaminophen hepatitis model is an experimental system that reproduces acute liver inflammation and injury caused by excessive acetaminophen exposure, enabling controlled study of hepatotoxicity. At high doses, hepatic metabolism generates the reactive metabolite N-acetyl-p-benzoquinone imine (NAPQI); when glutathione stores are depleted, NAPQI binds cellular proteins, promotes oxidative stress, and damages hepatocytes, triggering inflammatory responses. In medicine and biomedical research, this model helps evaluate liver injury mechanisms, biomarkers, and protective or therapeutic interventions while providing a standardized framework for studying drug-induced liver disease and assessing how treatments may preserve hepatic function.

An optical system was developed to visualize hepatic microcirculation with FITC-labeled erythrocytes and to measure the partial pressure of oxygen in the microvessels with laser-assisted phosphorimetry. This method can be used to investigate physiological and pathological mechanisms by analyzing microvascular structure, diameter, blood flow velocity, and oxygen tension.
