Activating Function

The activating function is a theoretical measure in bioelectricity that predicts where an applied electric field will excite or inhibit an elongated excitable cell, such as a nerve fiber. It is calculated from the second spatial derivative of the extracellular electrical potential along the fiber, indicating regions likely to depolarize or hyperpolarize when stimulation is applied. In medicine, this principle helps relate electrode placement, waveform, and tissue geometry to neural activation during spinal cord stimulation, deep brain stimulation, and neuroprosthetic development. By guiding computational models and experimental designs, the activating function supports more selective stimulation, improved treatment planning, and reduced unintended activation of nearby pathways.

Described here is a rapid and effective procedure for functional reconstitution of purified wild-type and mutant CFTR protein that preserves activity for this chloride channel, which is defective in Cystic Fibrosis. Iodide efflux from reconstituted proteoliposomes mediated by CFTR allows studies of channel activity and the effects of small molecules.

Functional Near-Infrared Spectroscopy for Recording Brain Activity During Social Interactions

2025

Source: Reindl, V., et al. Conducting Hyperscanning Experiments with Functional Near-Infrared Spectroscopy. J. Vis. Exp. (2019) This video demonstrates the use of functional near-infrared spectroscopy (fNIRS) to measure brain activity during social interactions. Participants engage in a coordinated task that increases activity in the prefrontal cortex, a region critical for social behavior, while the system detects changes in light absorption due to blood oxygenation. By analyzing the synchrony...

Drosophila In Vivo Calcium Imaging: A Method for Functional Imaging of Neuronal Activity

This video describes in vivo calcium imaging in Drosophila neurons using GCaMP. The featured protocol clip shows how to record changes in GCaMP fluorescence from mushroom body neurons during an olfactory conditioning experiment.

Measuring the Activation of Fc-Mediated Effector Functions by HA Antibodies

2025

This video demonstrates an assay measuring the antibody-mediated activation of T cell effector functions. Adding an influenza virus hemagglutinin (HA)-specific monoclonal antibody to transfected mammalian cells expressing HA results in the formation of immune complexes with HA. Upon introducing engineered T cells expressing Fc receptors and a luciferase reporter, the T cells are activated by the antibody-HA complexes, and this activation is detected by adding a luciferase substrate and...
