Apparent Permeability

Apparent permeability is a measure of how readily a compound crosses a biological barrier, making it an important indicator of drug absorption and transport in medicine. It is typically calculated from the rate of compound flux across a cultured cell monolayer, normalized to membrane area and the concentration difference between donor and receiver compartments; the result reflects passive diffusion, active transport, and barrier integrity. Researchers use apparent permeability assays with models such as intestinal epithelial or blood-brain barrier cells to compare drug candidates, predict oral or central nervous system exposure, and identify transport mechanisms that influence pharmacokinetics and therapeutic effectiveness.

We describe a live whole animal quantitative measurement for permeability of the embryonic zebrafish brain. The technique analyzes the ability to retain cerebrospinal fluid and molecules of different molecular weights within the neural tube lumen and quantifies their movement out of the ventricles. This method is useful for determining differences in epithelial permeability and maturation during development and disease.
