Casa System

The CASA system, or computer-assisted sperm analysis, is a laboratory method that uses digital microscopy and image-processing software to evaluate semen quality, supporting the assessment of male fertility. During analysis, a camera records sperm movement in a calibrated sample chamber, while software detects and tracks individual cells to quantify concentration, motility, velocity, and related motion patterns; some systems also assess morphology. In medicine, CASA provides objective, reproducible measurements that complement manual semen examination, helping clinicians investigate infertility, monitor changes during treatment, and assess samples used in assisted reproductive procedures. Standardized sample handling and instrument settings remain essential for reliable results.

An Enzymatic Method to Rescue Mesenchymal Stem Cells from Clotted Bone Marrow Samples

Mesenchymal stem cells are usually obtained from bone marrow and require expansion culture. When samples clot before processing, a protocol using the (enzymatic) thrombolytic drug urokinase can be applied to degrade the clot. Thus, cells are released and available for expansion culture. This protocol provides a rapid and inexpensive alternative to resampling.

Here we provide a protocol on how to package lentivector expression constructs in pseudoviral particles and to transduce target cells using the pseudoviral particles.
