Corneal Tissue Transplant

Corneal tissue transplant is a surgical procedure that replaces diseased or damaged corneal tissue with healthy donor tissue to restore vision and protect the eye. Depending on the location and extent of injury, surgeons may replace the full corneal thickness through penetrating keratoplasty or transplant selected layers using lamellar or endothelial techniques. The graft integrates with the recipient’s eye while preserving the cornea’s transparent structure, although rejection, infection, and healing complications require monitoring. Corneal transplantation can treat scarring, keratoconus, corneal swelling, and other conditions that impair vision, making it an important application of reconstructive ophthalmic medicine.

Corneal Donor Tissue Preparation for Endothelial Keratoplasty

Endothelial corneal transplantation is a surgical technique for treatment of posterior corneal diseases. Mechanical microkeratome dissection to prepare tissue results in thinner, more symmetric grafts with less endothelial cell loss and improved outcomes. Dissections can be performed at the eye bank prior to corneal transplantation surgery.

An all laser procedure is proposed in the endothelial transplant. The surgical technique is based on the use of a femtosecond laser to prepare the donor tissue. Laser welding technique is then used to secure the donor endothelium in the correct position.

Protocols for germ cell transplantation and testis tissue xenografting are described. Germ cell transplantation results in donor-derived spermatogenesis in recipient testes and represents a functional reconstitution assay for identification of spermatogonial stem cells (SSCs).
