Intrahepatic Biliary Cyst

An intrahepatic biliary cyst is a fluid-filled, cystic lesion arising within the liver and associated with the intrahepatic bile ducts, making accurate diagnosis important for distinguishing it from other hepatic cysts and biliary disorders. These lesions may result from congenital abnormalities of bile-duct development or localized duct dilation, allowing bile or clear fluid to accumulate within the liver. Ultrasound, computed tomography, and magnetic resonance cholangiopancreatography help characterize cyst location, internal features, and communication with the biliary tree. Clinical management depends on symptoms, infection, obstruction, growth, and malignant potential, with observation, drainage, or surgery considered in selected cases.

Robotic liver surgery has gained more acceptance as a feasible, safe, and effective procedure for the treatment of both benign and malignant indications. However, robotic left hepatectomy is still technically demanding. We describe our surgical technique of a robotic left hepatectomy using indocyanine green fluorescence imaging for a large biliary cyst.

Three-dimensional (3D) cellular systems are relevant models for investigating organogenesis. A hydrogel-based method for biliary cysts production and their characterization is proposed. This protocol unravels the barriers of 3D characterization, with a straightforward and reliable method to assess cyst formation efficiency, sizes, and to test their functionality.

To help assess the molecular mechanisms underlying zebrafish biliary-driven liver regeneration, we established a liver injury model in which the nitroreductase-expressing hepatocytes are genetically ablated upon metronidazole treatment. In this protocol, we describe how to adeptly manipulate, monitor and analyze hepatocyte ablation and biliary-driven liver regeneration.
