Lipolysis Quantification

Lipolysis quantification is the measurement of triglyceride breakdown and the release of fatty acids and glycerol from adipose tissue or other lipid-storing cells. It commonly relies on detecting glycerol or nonesterified fatty acids generated when lipases hydrolyze triglycerides, using biochemical assays, tissue sampling, or metabolic monitoring under defined conditions. In medicine, these measurements help assess adipose tissue function, energy metabolism, insulin resistance, obesity, and related metabolic disorders. They also support evaluation of hormones, drugs, and other interventions that alter fat mobilization, providing quantitative evidence for understanding metabolic regulation and developing targeted therapies.

This protocol describes the method of determining basal and forskolin-stimulated lipolysis in inguinal fat pads obtained from normal chow diet (NCD) or high fat diet (HFD) ± capsaicin fed wild type mice. As an index for lipolysis, glycerol release was measured from inguinal adipose fat pads.
