Melanoma Diagnosis

Melanoma diagnosis is the clinical process of identifying melanoma, a potentially aggressive skin cancer, and distinguishing it from benign or other malignant lesions. Evaluation typically begins with a skin examination and dermoscopy, which magnifies and illuminates lesions to reveal patterns of asymmetry, irregular borders, varied pigmentation, and structural changes; a biopsy then provides tissue for microscopic confirmation by a pathologist. Additional imaging or molecular testing may help assess disease extent and guide treatment in selected cases. Accurate diagnosis supports earlier intervention, informs staging and prognosis, and helps clinicians select appropriate surgical, systemic, or surveillance strategies.

This video describes the technique of isolating CD133 negative and positive melanoma stem cells from a primary melanoma cell line using magnetic-activated cell sorting. The CD133 positive melanoma CSCs are further studied to understand the mechanisms of melanoma tumorigenesis.

Screening for Melanoma Modifiers using a Zebrafish Autochthonous Tumor Model

A rapid way to screen for melanoma modifiers using a zebrafish autochthonous tumor model is presented. It takes advantage of the miniCoopR vector which allows for expression of candidate melanoma genes in melanocytes. A method to obtain melanoma-free survival curves, an invasion assay, a protocol for antibody staining of scale melanocytes and a melanoma transplantation assay are described.
