Murine Lung Transplant

Murine lung transplant is a surgical procedure that transfers lung tissue between mice to study graft function, immune responses, and mechanisms of transplant injury. In an orthotopic model, the donor lung is removed, preserved briefly, and implanted into the recipient, where surgeons reconnect the airway and pulmonary vessels to restore ventilation and blood flow. Researchers can vary donor and recipient genetics, immunosuppressive treatment, or experimental injury conditions to examine acute rejection, ischemia-reperfusion injury, and tolerance. Because the model permits controlled tissue sampling and longitudinal analysis, it supports investigation of transplant biology and evaluation of therapies relevant to human lung transplantation.

Obliterative bronchiolitis is the key impediment to the long-term survival of lung transplant recipients and the lack of a robust preclinical model precludes examining obliterative bronchiolitis immunopathogenesis. Unlike other solid organ transplants, vascularized mouse lung transplantation has only recently been developed. Here we show our independently developed obliterative bronchiolitis model after murine orthotopic single-lung transplantation.
