Myocardial Biopsy

Myocardial biopsy is an invasive diagnostic procedure that removes a small sample of heart muscle for microscopic and molecular evaluation, helping identify conditions that cannot be diagnosed reliably from imaging alone. During the procedure, a catheter, commonly introduced through a vein, guides biopsy forceps into a cardiac chamber to obtain endomyocardial tissue, which is then examined by histopathology, immunohistochemistry, or molecular testing. In medicine, the technique supports detection of myocarditis, cardiac amyloidosis, and other infiltrative disorders, and it is especially important for monitoring rejection after heart transplantation. Results can guide immunosuppressive or other targeted treatment while informing prognosis and further testing.

The post-mortem assessment of myocardial infarction (MI) in rodents is based on quantification of the infarct on stained heart sections. We describe an accurate method to quantify the infarct size using systematic sampling of harvested rat hearts from base to apex and image analyses of trichrome-stained histological sections.

This protocol describes the porcine myocardial infarction (MI) model using a 90 min closed-chest coronary balloon occlusion of the left anterior descending artery (LAD), followed by reperfusion.
