Serum Isolation

Serum isolation is a laboratory technique used to separate the liquid portion of clotted blood from blood cells and clotting components, providing a sample for medical analysis and research. During the process, whole blood is allowed to clot, and centrifugation separates the clear serum from the compact clot and cellular material; careful handling helps minimize hemolysis and contamination. The resulting serum contains proteins, antibodies, hormones, metabolites, and other soluble biomarkers. It supports diagnostic testing, disease monitoring, immunological studies, therapeutic research, and biochemical assays, making consistent collection and processing essential for reliable results.

This protocol describes a method for extracting small RNAs from human serum. We have used this method to isolate microRNAs from cancer serum for use in DNA arrays and also singleplex quantitative PCR. The protocol utilizes phenol and guanidinium thiocyanate reagents with modifications to yield high quality RNA.

Efforts to understand microglial function in detail have been hindered by the lack of microglial culture models that recapitulate the properties of mature in vivo microglia. This protocol describes an isolation and culture approach designed to maintain robust survival of highly ramified mature rat microglia under defined-medium conditions.

Development and Maintenance of Serum-Free Embryoid Bodies

2025

The video describes the procedure for developing serum-free embryoid bodies (SFEBs) from neuronal progenitor cells using various serum-free differentiation mediums or DM. DM-2 facilitates neurons' maturation and arrangement to form SFEBs, while DM-3 is used for further growth maintenance.
