Spared Nerve Injury Model

The spared nerve injury model is an experimental animal model of neuropathic pain in which injury to selected peripheral nerves produces persistent sensory abnormalities while another nerve remains intact. Typically, the tibial and common peroneal branches of the sciatic nerve are ligated or transected, leaving the sural nerve spared; this arrangement induces mechanical allodynia, hyperalgesia, and altered pain signaling in the affected limb. Researchers use the model to investigate peripheral nerve damage, neuroimmune interactions, and central sensitization, and to evaluate analgesic drugs and other interventions. Its reproducible behavioral and biological outcomes support studies of pain mechanisms and therapeutic development in medicine.

The Spared Nerve Injury animal model is described here as a mouse model of peripheral neuropathic pain following partial denervation of the sciatic nerve by lesioning the tibial and common peroneal nerve branches, leaving the remaining sural nerve intact. Behavioral modification resulting from mechanical allodynia is quantified by von Frey filaments.

Source: Svientek, S. R., et al. Fabrication of the Composite Regenerative Peripheral Nerve Interface (C-RPNI) in the Adult Rat. J. Vis. Exp. (2020).This video demonstrates the surgical construction of a composite regenerative peripheral nerve interface (C-RPNI) in a rat model of peripheral nerve injury. The procedure involves attaching a transected peroneal nerve to a muscle graft and overlaying it with a dermal skin graft to promote nerve regeneration. Over time, motor and sensory neurons...

Nerve-sparing Mid-urethral Obstruction (NeMO) in Female Small Rodents

Traditional modeling of partial bladder outlet obstruction in rodents is fraught with animal mortality. A denervation injury from dissection around the proximal urethra and bladder neck is also of major concern. We developed and evaluated a safe and reliable mid-urethral obstruction model, avoiding the shortcomings of the traditional model.
