Systolic Dysfunction

Systolic dysfunction is a condition in which the heart’s ventricles cannot contract effectively, reducing the amount of blood ejected with each heartbeat and potentially impairing tissue perfusion. It commonly results from weakened or damaged cardiac muscle, which lowers stroke volume and ejection fraction and can activate compensatory pathways that increase heart rate, fluid retention, and vascular resistance. In medicine, assessing systolic function with echocardiography and related tests helps identify heart failure with reduced ejection fraction, evaluate causes such as myocardial infarction or cardiomyopathy, and guide treatment. Recognizing the underlying mechanism supports therapies that reduce symptoms, limit ventricular remodeling, and improve clinical outcomes.

We describe the creation of a rat model of pressure overload induced moderate remodeling and early systolic dysfunction where signal transduction pathways involved in the initiation of the remodeling process are activated. This animal model will aid in identifying molecular targets for applying early therapeutic anti-remodeling strategies for heart failure.

Early stage hemodynamic dysfunction is critical to the development of kidney disease. Yet, detection methodologies are limited. Recent advances in sonography provide a noninvasive, accurate option for early detection of kidney injury. This study outlines a step-by-step, sonographic methodology for detecting kidney dysfunction using a drug-induced nephrotoxicity rat model.

Mouse model of metabolic dysfunction-associated steatotic liver disease (MASLD) with metabolic dysfunction, hepatic gene expression changes, and liver histopathological alterations that resemble human MASLD, including fibrosis that progresses to advanced fibrosis stage 3. This model can be used in studies of MASLD pathophysiology and in pre-clinical studies of new therapies.

Here, we present a protocol to enable the capture and identification of disease-specific protein-protein interactions from native cells and tissues using chemical probes and mass spectrometry. The resulting interaction datasets are analyzed through a dedicated web-based platform to reveal dynamic network dysfunctions and pathway alterations linked to disease.
