Vaccine Efficacy Evaluation

Vaccine efficacy evaluation is the systematic assessment of how well a vaccine prevents infection, disease, or other defined outcomes under controlled conditions, making it central to evidence-based medicine. In randomized controlled trials, researchers compare outcome rates in vaccinated and placebo or control groups, then calculate efficacy from the relative reduction in risk while monitoring safety and immune responses. Follow-up data can show protection against symptomatic illness, severe disease, or infection across populations and time periods. These evaluations guide regulatory decisions, vaccination recommendations, clinical practice, and later effectiveness studies that measure performance in routine healthcare settings.

The determination of colony-forming units (CFU) is the gold-standard technique for quantifying bacteria, including Mycobacterium tuberculosis which can take weeks to form visible colonies. Here we describe a micro-CFU for CFU determination with increased time efficiency, reduced lab space and reagent cost, and scalability to medium and high throughput experiments.

In this video, we describe a protocol to test the efficacy of an acellular pertussis vaccine against Bordetella pertussis infection in mice. Post-vaccination, the mice were subjected to infection, and their tissue homogenate was cultured to analyze the bacterial colonies as a readout of the vaccine-induced protective immunity.
