Automated Behavioral Assay

An automated behavioral assay is a method that uses sensors, cameras, and software to measure and analyze animal or human behavior with limited observer intervention. In neuroscience, these systems capture responses to controlled stimuli and apply computer vision, motion tracking, or signal-processing algorithms to quantify behaviors such as locomotion, exploration, learning, and social interaction. Automated data collection improves measurement consistency, temporal resolution, and experimental throughput while reducing observer bias. These assays support studies of neural circuits, brain disorders, drug effects, and genetic influences, helping researchers connect behavioral changes with underlying biological mechanisms.

This step-by-step protocol analyzes Drosophila negative geotaxis behavior using an automated multi-cylinder system that hosts hundreds of flies and synchronizes their action by an electric motor. Upon synchronization, fly negative geotaxis behavior is assayed, digitally recorded, and analyzed using the self-designed RflyDetection software.

Our laboratory developed a novel high-throughput automated imaging system that is useful for the detection of several different behaviors in 7-day-old zebrafish larvae. The system can be used for detecting subtle changes in behavior after the larvae have been exposed to environmental toxicants or pharmaceuticals.

Automated Analysis of C. elegans Swim Behavior Using CeleST Software

An efficient and simple methodology for computer-based analysis of nematode swimming behavior in liquid is described. The method requires little to no investment for C. elegans laboratories. The hardware used is standard, and the computer software for the behavioral analysis (CeleST) is an open source one.
