Dendritic Arbor Imaging

Dendritic arbor imaging is the visualization and quantitative analysis of a neuron's branched dendritic structure, which receives and integrates signals from other cells. By labeling dendrites and capturing high-resolution microscopy images, researchers can reconstruct arbor geometry and measure features such as branch length, branching patterns, and spine distribution. These measurements reveal how neuronal architecture develops, adapts during synaptic plasticity, and differs across brain regions or disease conditions. In neuroscience, dendritic arbor imaging links cellular structure to function and supports studies of circuit formation, experience-dependent remodeling, and disorders associated with altered neuronal connectivity.

We describe two methods for visualization and quantification of dendritic arborization in the hippocampus of mouse models: real-time and extended depth of field imaging. While the former method allows sophisticated topographical tracing and quantification of the extent of branching, the latter allows speedy visualization of the dendritic tree.

Source: Gouder, et. al. Three-dimensional Quantification of Dendritic Spines from Pyramidal Neurons Derived from Human Induced Pluripotent Stem Cells. J. Vis. Exp. (2015).This video demonstrates the imaging and analysis of dendritic spines in transduced, immunolabeled pyramidal neurons using confocal microscopy. Background noise is reduced, and dendrites are traced by estimating their diameter and length. Automated spine segmentation involves setting parameters for dendrite and spine dimensions...

Here, we present a method for investigating neurite morphogenesis in postnatal mouse retinal explants by time-lapse confocal microscopy. We describe an approach for sparse labeling and acquisition of retinal cell types and their fine processes using recombinant adeno-associated virus vectors that express membrane-targeted fluorescent proteins in a Cre-dependent manner.
