En Bloc Tissue Retrieval

En bloc tissue retrieval is a method for removing a tissue or organ region as an intact unit, preserving spatial relationships that can be lost when samples are collected piecemeal. In neuroscience, the specimen is carefully dissected from surrounding structures, stabilized through handling and fixation, and then processed for sectioning, staining, imaging, or molecular analysis. Preserving continuous anatomy helps researchers examine neural circuits, lesions, interfaces between brain regions, and pathological changes within their original context. The approach supports accurate histological mapping and can improve interpretation of structural relationships in developmental, experimental, and neuropathological studies.

En Face Preparation of Mouse Blood Vessels

A procedure for making en face preparations of the mouse carotid artery and aorta is described. Such preparations, when immunofluorescently stained with specific antibodies, enable us to study localization of proteins and identification of cell types within the entire vascular wall by confocal microscopy.

We describe a technique for the preparation of clarified human cortical homogenates, protein separation by SDS-PAGE, antigen retrieval and immunoblotting with an antibody to the Aβ peptide. Using this protocol, we consistently detect monomeric and multimeric Aβ in cortical tissue from humans with Alzheimer's pathology.
