Ethanol Sedation Assay

The Ethanol Sedation Assay is a behavioral test that measures how ethanol-induced central nervous system depression affects an animal’s loss and recovery of the righting reflex. After ethanol administration, researchers record the time required for an animal to lose its ability to turn upright and the duration until normal posture returns, providing a quantifiable measure of sedation sensitivity. In neuroscience, the assay helps evaluate ethanol responses across strains, developmental stages, or experimental treatments and can reveal changes in inhibitory signaling, ethanol tolerance, and genetic or pharmacological factors that influence alcohol-related behavior. These outcomes support studies of neural circuits involved in intoxication and recovery.

Microdialysis of Ethanol During Operant Ethanol Self-administration and Ethanol Determination by Gas Chromatography

A method to determine the time course of ethanol concentration in the brains of rats during operant ethanol self-administration is described. Gas chromatography with flame ionization detection is used to quantify ethanol in the dialysate samples, because it has the sensitivity required for the small volumes that are generated.

C. elegans is a useful model for studying the effects of ethanol on behavior. We present a behavioral assay that quantifies the effects of ethanol on the locomotion speed of crawling worms; both initial sensitivity and the development of acute functional tolerance to ethanol can be measured with this assay.
