Ferret Brain Tissue

Ferret brain tissue is the neural tissue of ferrets, including interconnected neurons, glial cells, blood vessels, and supporting extracellular structures that enable sensory processing, cognition, and coordinated behavior. Its functions arise through electrical signaling between neurons, chemical communication at synapses, and regulated interactions between neural and vascular cells. In neuroscience, ferret brain tissue provides a valuable model for studying cortical organization, visual system development, neural circuits, and responses to injury or disease. Researchers examine intact tissue, tissue sections, and cultured cells using anatomical, electrophysiological, molecular, and imaging methods to connect cellular structure with brain function.

The method describes inflammation-sensitized hypoxic-ischemic and hyperoxic brain injury in the P17 ferret to model the complex interaction between prolonged inflammation and oxidative brain injury experienced in a number of late preterm infants.

Frozen Mouse Brain Tissue Sectioning: A Procedure to Obtain Thin Frozen Tissue Sections from Frozen Murine Brain Tissue

2023

In this video, we demonstrate the sectioning of a frozen mouse brain tumor tissue using a cryostat. The frozen brain tissue sections so obtained are stored at low temperatures until further analysis.

Source: Kirchner, K.N., et al., A Hydrophobic Tissue Clearing Method for Rat Brain Tissue. J. Vis. Exp. (2020).This video demonstrates the method of hydrophobic tissue clearing of a rat brain tissue sample. The sample is sequentially dehydrated in methanol, treated with a dichloromethane and methanol mixture to dissolve lipids and pigments, and transferred to dibenzyl ether for complete tissue clearing, enabling detailed imaging without thin sectioning.

We describe a protocol for transcardiac perfusion of mice, removal and sectioning of the brain, as well as immunoperoxidase staining, resin embedding, and ultrathin sectioning of the brain sections. Upon completion of these procedures, the immunostained material is ready for examination with transmission electron microscopy.
